1) circRNA-seq
Total RNA去除rRNA后，采用Rnase R去除线性RNA，对富集得到的circRNA进行打断处理并建库测序。

a) 降解线性RNA，特异性高；
b) 能够检测较全的circRNA；
2) 全转录组RNA-seq
全转录组RNA-seq采用链特异性建库策略，既可检测mRNA和lncRNA的表达，还可同时对circRNA进行检测，并对三者进行联合分析。

十、 样品要求
A. 样本类型：细胞、新鲜正常或疾病组织、total RNA；
B. 样品量：circRNA-seq：total RNA≥ 20µg；全转录组测序：total RNA≥ 5µg；
C. 样本质量：OD260/280值应在1.9～2.2之间；DNA应该去除干净，RNA无降解；RIN值≥7；
D. 样品保存运输：-80℃冰箱或者液氮冻存，干冰运输。
